纯化蝎毒素Ⅳ对神经损伤康复的促进作用

摘    要:实验用大鼠20只,结扎坐骨神经以造成神经慢性机械压迫-痛觉过敏动物模型。实验组从手术后第2天开始经腹腔注射纯化蝎毒素Ⅳ,每日注射600mg/kg,持续用药,选用辐射热测痛.本实验用电生理学方法结合HRP逆行追踪技术进行研究。结果显示,纯化蝎毒素Ⅳ对神经损伤后形态和功能的恢复以及对神经损伤性痛觉过敏的解除都具有促进作用.

Abstract:rats were divided into experiment and control groups, experiments were carried out in five steps as below: (1) sciatic nerve on right side were ligated: (2) 2 days after operation, experiment group animals were treated with SVC Ⅳ (600 mg/kg/d) through abdominal injection; (3) the latencies of withdrawal reflex of the hind paws were measured by the apparatus of radiant heat before and 7 d, 13 d, 20 d, after operation ; (4) 20 days after operation, bilateral nervi saphenus were transected, the right lateral gastrocnemius were placed on recording electrode, the right paw pad were placed on stimulating electrode, the threshold of the flexion reflex was observed ; (5) HRP was injected into bilateral gastrocnemius and bilateral hypodermis of the paw pad, the labeled cells in the ganglion spinale (L4-L6) and cornu ventrale of spinal cord(L4-L6) were checked. Results: (1) 20 days after operation, the latencies of withdrawal reflex in experiment group was much longer than that in control group; (2) on 6± 0. 8 V stimulus, the flexion reflex of experiment group was evoked by both A-afferent fibers and C-afferent fibers whereas only C-afferent fibers activities appeared in control group,on 10 V stimulus; (3) experiment group, the percentage of restoring neurons in the ganglion spinales was 28. 2±7. 3 and in the cornu ventrale was 21. 1± 7. 7; in control group, it was 14. 2± 5. 6 and 5. 8±2. 8 respectively.Our experiments proved SVC Ⅳ might promote the restoration of peripheral nerve injury and related principles were discussed.
