硒蛋白K在小鼠中的组织差异表达及T淋巴细胞内定位

摘    要:目的检测硒蛋白K(Sel K)在小鼠脾、肾、肠、肺、脑、肝组织中的表达差异及在T淋巴细胞内的定位,为研究Sel K的分布与生物学功能的关联性提供参考。方法采用实时荧光定量PCR,Western blot以及免疫组化方法检测硒蛋白K在小鼠各组织的表达差异,并采用细胞免疫化学的方法 ,用激光共聚焦显微镜观察硒蛋白K在小鼠T淋巴细胞内的定位。结果实时荧光定量PCR结果显示硒蛋白K在小鼠各组织中均有表达,并且在脾脏中的表达明显高于其他组织(P0.05)。Western blot及免疫组化结果与实时荧光定量PCR结果基本一致。细胞免疫化学结果显示硒蛋K在小鼠T淋巴细胞中定位于内质网。结论硒蛋白K是一种在脾脏中高表达的内质网蛋白。

Expressions of selenoprotein K in tissues and subcellular localization in T lymphocyte of mice

Abstract:Objective To determine the expression of selenoprotein K (SelK) in the spleen, kidney, intestine, lung, brain tissues and subcellular localization in T lymphocyte of mice ,for establishing connections between functions and tissue distribution or subcellular localization of SeIK. Methods The expression of selenoprotein K(SeIK) was detected in the different tissues of mice by real time RT-PCR, Western blot and immunohistochemical. Subcellular localization of SelK in T lymphocyte was detected by immunocytochemistry/immunofluorescence. Results The real time RT-PCR result showed that the expression level of SelK in spleen was significantly higher than other tissues (P〈0.05). The results of Western blotting and immunohistochemical were basically consistent with the real time RT-PCR result. ICC/IF results showed that SelK was located in the endoplasmic reticulum of T lymphocytes. Conclusion SelK is an endoplasmic reticulum protein with higher expression level in spleen than other tissues.
