PPARγ基因静默抑制裸鼠肝癌肺转移

摘    要:目的探讨过氧化物酶体增殖物活化受体γ(Peroxisome proliferator-activated receptor gamma,PPAR-γ)基因静默对肝癌裸鼠模型肺转移的影响。方法构建PPARγ短发夹状RNA表达质粒并转染HCCLM3细胞(pshPPARγ组),以空质粒转染为对照组。观察两组皮下种植瘤生长曲线及体积,肺转移灶数量及分级。结果pshPPARγ组种植瘤体积为(1.86±0.65)cm^3,肺转移灶数量为(37.2±0.7)个/肺,分级为1—2级;对照组体积为(4.86±1.15)cm^3,肺转移灶数量为(107.8±6.1)个/肺,分级多为3—4级,两组种植瘤体积和转移灶数量、分级的差异有统计学意义(P〈0.05)。结论PPARγ基因静默可降低肝癌种植瘤肺转移数量。

Abstract:Objective To investigate the effect on pulmonary metastasis of hepatocellular carcinoma in nude mice with short hairpin RNA (shRNA) targeting at peroxisome proliferator-activated receptor gamma (PPAR γ) gene.Method shRNA targeting at PPAR γ gene was constructed and transfected into HCCLM3 cells (pshPPARγ group), meanwhile, cells transfected with vector plasmids as control group.Positive clones were selected and constructed transplanted tumor animal models in nude mice; growth and volume of tumors were observed. After 8 weeks, PPAR γ mRNA and protein in transplanted tumor tissues were detected, pulmonary metastases were counted and graded.Result PPARγ mRNA down regulated significantly in pshPPAR γ group, its inhibition rate is 82%. In pshPPAR γ group, tumor volume was (1.86 ± 0.65)cm^3, pulmonary metastases were at grade 1-2 and 37.2 ± 0.7 per lung; while in control group, tumor volume was (4.86± 1.15)cm^3, pulmonary metastases were at grade 3-4 and 107.8 ± 6.1 per lung.There are significant differences comparing with control group (P〈0.05). Conclusion RNA interference targeting at PPARγ gene can impair pulmonary metastasis of HCC in nude mice.
