红丝线草对胰岛素抵抗脂肪肝大鼠有关指标的影响

目的:观察红丝线草提取物(Peristrophe roxburghiana,HSX)对胰岛素抵抗脂肪肝大鼠胰岛素敏感性、血脂和肝脏功能的影响。方法:采用高脂高糖饮食建立胰岛素抵抗脂肪肝动物模型,并连续8周预防性灌胃给予HSX处理。8周后测定糖耐量,处死大鼠,取血测定FFA、TC、TG、HDL-C、LDL-C、AST、ALT、FBG和FINS,并计算ISI;取全肝称重,计算肝指数;取肝左叶相同部位组织,常规HE染色,光镜下评估肝组织脂肪变性的程度。结果:HSX高、低剂量处理后,模型组大鼠空腹血清胰岛素,血浆FFA、TG水平基本恢复正常,并提高血浆HDL-C水平,降低ALT和AST活性,降低肝指数,防止肝组织脂肪性病变;同时,低剂量的HSX可增加胰岛素敏感性,改善糖耐量。结论:HSX对高脂高糖饮食引起的胰岛素抵抗脂肪肝大鼠具有调脂保肝作用,其作用可能与提高胰岛素敏感性有关。

Effect of Peristrophe Roxburghiane on Insulin Sensitivity ,Serum Lipid and Liver Function in Rats of Insulin Resistance with Fatty Liver

Objective:To observe the effects of Peristrophe roxburghiane on insulin sensitivity,serum lipid,liver function in rats of insulin resistance with fatty liver.Methods:Adult SD rats were fed with high-fatsugar diet,then peristrophe roxburghiane was administered to the rats for 8 weeks.And then the levels of FFA,TC,TG,HDL-C,LDL-C in blood sample,the activities of AST,ALT,fasting serum glucose(FBG)and insulin were measured,and insulin-sensitivity index was calculated.The liver was weighed and collected to observe the histologica changes,and liver index was calculated.Results:Treatment of peristrophe roxburghiane can decrease the levels of FFA and TG,increase HDL-C,lower the activities of ALT and AST,and improve the histological changes of liver tissues.Low dose of peristrophe roxburghiane can elevate insulin sensitivity and improve glucose tolerance.Conclusions:Peristrophe roxburghiane possesses the regulation of serum lipid and protection of the liver function on the rats of insulin resistance with fatty liver,and these actions may be concerned with improvement of the insulin sensitivity.
