小鼠腹部异位心脏移植模型的建立与改进

摘    要:【】目的 建立并改进小鼠腹部异位心脏移植模型,为研究器官移植免疫学提供支持。方法 结合国内外文献报道的手术方法,将供者心脏的升主动脉和肺动脉分别与受者腹主动脉和下腔静脉行端侧吻合,建立小鼠异位心脏移植模型,并在传统方法的基础上对受体准备和供体心脏摘取、修整以及血管吻合技术加以改良,观察改进后的手术效果并分析其优势。结果 预实验完成50对。正式实验同系间移植完成20对,成功17对,成功率为85%。同种间移植完成15对,成功13对,成功率为86.7%。模型总成功率为85.7%。受体血管吻合前准备时间平均为11.2 ± 2.5分钟,供体心脏摘取、修整时间平均为13.6 ± 3.3分钟,供、受体血管吻合时间平均为21.7 ± 3.5分钟。改良后的心脏移植模型简化了手术操作,降低了手术难度。结论 改进的小鼠腹部异位心脏移植模型具有简便、成功率高等优势,为进一步进行移植免疫学研究奠定基础。

Abstract:Objective To establish and improve heterotopic abdominal heart transplantation in mice and support for research in organ transplantation immunology. Methods After widely reading the literature, we established a mouse model of heterotopic heart transplantation by suturing the donor ascending aorta and the pulmonary artery to recipient abdominal aorta and inferior vena cava, respectively. Furthermore, we improved the preparing for recipient, extracting and preparing donor heart, and suture technology, observed the result, and analyzed the advantage. Results Preliminary experiments were between 50 couples mice. For formal experiments, syngeneic cardiac transplantation was performed between 20 couples and the success rate was 85%. Simultaneously, allogeneic cardiac transplantation was performed between 15 couples and the success rate was 86.7%. The total success rate was 85.7%. Preparing recipient time was 11.2 ± 2.5 min, extracting and preparing heart graft time was 13.6 ± 3.3 min, suturing time 21.7 ± 3.5 min. The improved cardiac transplantation technology simplified operation and declined difficulty. Conclusion The improved transplantation model possesses the advantage of simpler and higher success rate.
