摘    要:目的:探讨共济失调性毛细血管扩张症突变基因(ATM)mRNA在乳腺癌组织中的表达及其临床意义.方法:用原位分子杂交方法检测56例乳腺浸润性导管癌(BIDC)、37例导管原位癌(BDCIS)及53例癌旁正常组织(NBTAC)中的ATM mRNA的表达.结果:①ATM mRNA在BIDC中的表达明显低于NBTAC(P< 0.01).②ATM mRNA在BDCIS的表达低于NBTAC,但无统计学意义.③ATM mRNA在Ⅲ级BIDC中的表达率明显低于Ⅰ级分化组(P<0.05),但Ⅰ与Ⅱ级和Ⅱ与Ⅲ级分化组间ATMmRNA表达率差别无统计学意义.④ATM mRNA表达与肿瘤组织分化程度间呈正相关(r=0.312,P<0.05),即组织分化越低,ATM mRNA表达越低.⑤ATM mRNA的表达与淋巴结转移与否无关.⑥ATM mRNA表达在患者临床分期Ⅱ+Ⅲ期中的表达明显低于Ⅰ期的表达(P<0.05).结论:ATM mRNA在BTDC组织中表达降低,可为乳腺癌的早期发现和基因干预治疗提供新的思路.

Abstract:Objective: To investigate the expression and clinical significance of ataxia-telangiectasia mutant mRNA in breast carcinoma.Methods: The expression of ATM mRNA in 56 cases of breast invasive ductal carcinoma(BIOC),37 cases of breast ductal carcinoma in situ(BDCIS)and 53 cases normal breast tissues adjacent to carcinoma(NBTAC)were detected by in situ hybridization.Results: The expression rate of ATM mRNA in BIDC tissues was significantly lower than NBTAC(P<0.01).The expression rate of ATM mRNA in BDCIS was lower than NBTAC,but there was no significance.The expression rate of ATM mRNA in BIDC tissues Ⅲ grade was significantly lower than the Ⅰ grade(P<0.05),but there was no difference between the Ⅰ and Ⅱ grade or Ⅱ and Ⅲ grade.The expressions of ATM mRNA was positively correlated with the differentiation of BIDC(r=0.312,P<0.05).No significant correlation was observed between ATM mRNA expression and patients in lymphnode metastasis.There was significant difference between the clinic pathological manifestations of BIDC(P<0.05).Ⅱ and Ⅲ stage was lower than Ⅰ stage.Conclusion: The reduced expression of ATM mRNA in breast carcinoma maybe provide the new thought for the early detection and gene therapy of breast cancer.
