雄黄对HL—60／ADR作用的初步研究

目的：探讨中药雄黄对HL－60／ADR耐药细胞在抑制生长、逆转耐药、诱导凋亡方面的作用和影响。方法：（１）MTT比色法观察对细胞的换制；（2）用免疫组化法检测细胞Pgp的变化；（3）流式细胞仪检测细胞凋亡率。结果：（1）雄黄抑制HL－60／ADR耐药细胞的生长，药物浓度与HL－60／ADR细胞生长抑制率之间呈指数关系；（2）随着药物浓度的增加和时间的推移，HL－60／ADR细胞中MDR基因的表达产物Pgp阳性率逐步降低；（3）随着药物浓度的增加，HL－60／ADR细胞凋亡率增加。结论：雄黄可抑制HL－60／ADR 细胞生长，逆转其耐药，促进细胞凋亡，与剂量呈依赖性，其机制可能与Pgp表达下降有关。

STUDIES ON THE REVERSE OF RESISTANCE USING ARSENIC SULFIDE IN HL-60/ADR CELLS

Objective:To study the effect of arsenic sulfide on the growing, the reverse and the apoptosis of the HL-60/ADR cells.Methods:(1) Analyse the growth inhibition of the cells by MTT assay.(2)Test the Pgp by the means of immunohistochemistry . (3)Detect the cell apoptosis by flow cytometry. Results:(1) Treatment of HL-60/ADR cells with arsenic sulfide inhibited the cell growth in a dose-dependent manner. (2)Positive findings of Pgp decreased after incubation with drugs. (3)The percentage of apoptotic HL-60/ADR cells increased after incubation with drugs.Conclusion:Arsenic sulfide can inhibit the HL-60/ADR cell growth, reverse the resistance and induce the apoptosis in a dose- and time-depentent manner, while positive findings of Pgp decreased.
