钙离子结合蛋白A4和基质金属蛋白酶2在胰腺癌中的表达与浸润转移和预后的关系

摘    要:目的研究钙离子结合蛋白A4(S100A4)、基质金属蛋白酶2(MMP2)在胰腺癌组织中的表达与临床病理因素及其浸润、转移和预后的关系。方法采用SP免疫组化法检测82例胰腺癌、43例胰岛素细胞瘤和21例正常胰腺组织中S100A4和MMP2的表达情况。结果S100A4、MMP2在胰腺癌组织中的阳性表达率分别为69.51%(57/82)和58.54%(48/82);S100A4、MMP2蛋白在胰腺癌癌组织中表达明显高于正常组织和腺瘤(P〈0.05),S100A4、MMP2蛋白的表达与有无淋巴结转移、肿瘤的大小、TNM分期、分化程度以及CA199的值有关(P〈0.05);两种蛋白在胰腺癌组织中的表达呈正相关(P〈0.05)。S100A4和MMP2的表达情况均对患者的生存期有显著影响(P〈0.05)。结论S100A4、MMP2在肿瘤侵袭转移中发挥了重要作用,检测S100A4和MMP2的表达可作为预测胰腺癌的转移趋势指标,并能有效的评估胰腺癌患者的预后。

Abstract:Objective To observe the expressions of metastasis - associated protein ( S100A4 ) and matrix metalloproteinase 2 ( MMP2 ) in pancreatic cancer, insulinoma and normal pancreas, and investigate their correlations with the infiltration, metastasis and prognosis of pancreatic cancer. Methods The expression of S100A4 and MMP2 proteins was detected by immunohistochemistry with SP method in 82 specimens of pancreatic cancer,43 specimens of insulinoma and 21 normal tissue specimens. Results The positive rates of S100A4 and MMP -2 were 69.51% and 58.54% in pancreatic cancer tissue. The expression of S100A4 and MMP2 in the tissue of pancreatic cancer was significantly higher than that in insulinoma and normal pancreas (P 〈0.05). The expression of S100A4 and MMP2 protein was associated with tumor size,lymphatic metastasis, TNM stages, pathological types and CA199 quantitative value (P 〈 0.05). The expression of S100A4 and MMP2 protein were positive correlated with pancreatic cancer ( P 〈 0.05 ) . Univariate survival analysis confirmed expressions of S100A4 and MMP2 on survival time of pancreatic cancer patients (P 〈 0.05 ). Conclusions S100A4 and MMP - 2 proteins play a critical role in tumor metastasis. Co - detection of the expression of S100A4 and MMP - 2 can be used to predict the metastatic potential of pancreatic cancer, and can effectively evaluate the prognosis of patients with pancreatic cancer.
