山莨菪碱舒张微血管作用与血管内皮的关系

摘    要:目的 研究山莨菪碱舒张微血管作用与内皮细胞的关系及其可能的作用机制。方法 以去甲肾上腺素预收缩大鼠肠系膜血管床后给予山莨菪碱 ,观察去除血管内皮、给予NO合酶抑制剂左旋硝基精氨酸甲酯 (L NAME)、环氧酶抑制剂吲哚美辛和ATP敏感性钾通道 (KATP)开放抑制剂格列本脲后对血管张力变化的影响。结果 山莨菪碱对肠系膜血管床的舒张作用在去除血管内皮后 ,以 1μmol·L- 1的吲哚美辛、10 μmol·L- 1的L NAME或 1μmol·L- 1的格列本脲预处理后均显著减弱。结论 山莨菪碱的舒张微血管作用有内皮依赖性 ,且与内皮源性舒张因子、前列环素和KATP开放均有关

Abstract:AIM To study relationship between vasodilation effects of anisodamine and endothelium of microvessels. METHODS Norepine-phrine precontracted mesenteric microvascular beds in rats were used to determine changes in tension of vessels in response to anisodamine after removal of endothelium, coincubated with NG-nitro-L-arginine methyl ester(L-NAME), nitric oxide synthase inhibitor, indomethacin, cyclo- oxygenase inhibitor and glibenclamide, ATP-dependent K+ channels(KATP) inhibitor. RESULTS Anisodamine produced a vasodilation effect on mesenteric vascular beds, which was remarkably inhibited after rem oval of endothelium (P<0.01). The vasodilation effect of anisoda mine was also significantly inhibited after preconditioning with indomethacin(1 μmol·L-1), L-NAME (100 μmol·L-1) and glibenclamide (1 μmol·L-1) (P<0.05, P<0.01). CONCLUSION Anisodamine induced an endothelium dependent vasodilation which related to endothelium derived relaxing factor, prostacyclin and opening of KATP.
