摘    要:目的建立离体多巴胺(dopamin,DA)能神经细胞系MN9D细胞损伤模型。方法离体细胞培养条件下,于培养液中加入不同浓度6-羟基多巴胺(6-hydroxydopamine,6-OHDA)作用于MN9D细胞30 min。24 h后,MTT比色法检测MN9D细胞的存活率、Hoechst 33258核染色和流式细胞仪检测MN9D细胞的凋亡变化。结果6-OHDA以浓度依赖性的方式引起MN9D细胞的凋亡,导致细胞存活率降低。200μmol/L浓度的6-OH-DA作用30 min、培养24 h后,MN9D细胞的存活率降低至68.8%左右,凋亡率增高至约37.8%。结论6-OH-DA能够引起MN9D细胞的存活率降低,而凋亡率增高,产生类似帕金森病时的DA能神经细胞损伤的表现。

Abstract:Objective To establish the cell injury model of dopaminergic cell line MN9D in vitro.Methods MN9D cells were exposed to different concentrations of 6-hydroxydopamine(6-OHDA) for 30 min in vitro,and 24 h later,the survival of MN9D cells was detected with MTT chromatometry,and the MN9D cells apoptosis was determined with Hoechst 33258 staining and flow cytometry.Results 6-OHDA caused the apoptosis of MN9D cells in a concentration-dependent manner: the survival rate of cells decreased approximately to 68.8% while the apoptotic percentage of cells increased to 37.8% 24 h after a 30 min exposure to 200 μmol/L 6-OHDA.Conclusion 6-OHDA could induce an decrease in the survival rate of MN9D cells and an increase in their apoptosis percentage,causing an injury to MN9D cells in vivo in a form similar to the injury model of dopaminergic neurons in Parkinson′s disease(PD).
