白细胞介素13基因-1112C/T多态性与哮喘、血浆细胞因子水平的相关研究

摘    要:目的探讨血浆IgE、IL-13水平在哮喘和对照组之间的差异以及IL-13基因-1112位点具有相同基因型的个体二者的相关性。方法用聚合酶链反应和限制性片段长度多态性(PCR/RFLP)方法检测哮喘与对照组-1112位点多态性,用酶联免疫吸附法(ELISA)测定血浆IgE、IL-13水平。结果IL-13基因-1112位点存在3种基因型。在哮喘和对照组,具有相同基因型的个体,其细胞因子水平存在显著性差异,P<0.01。在哮喘或对照组,具有相同基因型的个体,其IgE及IL-13水平存在显著性正相关,r=0.63~0.74,P<0.01。结论IgE、IL-13是影响哮喘发病的重要细胞因子,且二者存在显著性正相关。

Abstract:Objective: To investigate the differences of total plasma IgE and IL-13 levels between asthma and control group,and the correlation between total plasma IgE and IL-13 levels with the same genetype in -1112 site polymorphism of IL-13 gene. Methods: Polymerase chain reaction/restriction fragment length polymorphism (PCR/RFLP) techniques were used to determine -1112 locus polymorphism in asthma and control group,and total plasm IgE,IL-13 levels were determined by ELISA . Results: There are three genetypes in -1112 locus. The cytokine levels of the subjects with the same genetype have significant difference (P < 0.01) between asthma and control groups. The total plasma IgE levels were positively correlated with the IL-13 levels of the ones with the same genetype ( r=0.63~0.74,P < 0.01) in either asthma or control group. Conclusion: IgE and IL-13 play important roles in asthma,and one positively correlates with another.
