摘    要:目的:探讨更为简单,廉价的人星形细胞瘤细胞原代培养方法。方法:应用机械分散法,对不同级别的新鲜星形细胞瘤手术标本进行肿瘤细胞原代培养及传代培养,通过调整培养参数,观察肿瘤细胞的生长情况,并从形态学及细胞免疫组化染色等方法进行研究。结果:Ⅱ级及以上的星形细胞瘤均成功进行原代细胞培养,Ⅲ及Ⅳ级星形细胞瘤还成功获得传代培养,并表达免疫组化GFAP,不表达Ⅳ胶原。结论:星形细胞瘤细胞的原代培养可以用机械分散法成功培养,该方法简单易操作,重复性好且价格低廉,可以较好的满足实验要求。

Abstract:Objective : To find an method of primary culture of astrocytoma cell in vitro. Methods : Using mechanical dispersion method, astrocytes from the human astrocytoma tissues were cultured and identified by immunohistochemistry identification. Results: Primary glioma were successfully cultured and immunohistoehemistry analysis showed that the cells were stained positive for GFAP. Conclusion: The method of mechanical dispersion can successfully culture primary astroeytoma. This method is simple and easy-to-use, inexpensive and reproducible, and can better meet the re- quirements of the experiment.
