膀胱癌中Twist蛋白表达与上皮-间质转化的关系

目的 探讨膀胱癌中上皮-间质转化因子Twist表达及其与上皮-间质转化及病理参数之间的关系.方法 应用E-钙黏蛋白和N-钙黏蛋白的表达来检测上皮-间质转化的发生.采用免疫组织化学SABC法、实时定量聚合酶链反应(PCR)和Western blot法观察45例膀胱癌组织和5例正常膀胱组织中Twist蛋白、E-钙黏蛋白和N-钙黏蛋白的表达.结果 45例膀胱癌组织和5例正常膀胱黏膜中Twist阳性率分别为26.66%和0,随着膀胱癌的病理分级及临床分期的增高,Twist的表达率逐渐升高.且Twist在有淋巴转移中的表达明显高于无淋巴结转移的膀胱癌.Twist的表达与E-钙黏蛋白和N-钙黏蛋白的表达显著相关.结论 Twist蛋白的表达对上皮-间质转化有重要影响,可为膀胱癌侵袭转移的研究和患者的预后判断提供参考.

Relationship between epithelial-mesenchymal transition and Twist expression in bladder cancer

Objective To investigate the relaitonship between the immunohistochemical expression of Twist and epithelial-mesenchymal transition in bladder cancer. Methods The epithelial-mesenchymal transition was assessed by the expression of E-cadherin and N-cadherin. The expression of E-cad-herin and N-cadherin in formalin-fixed and paraffin-embedded tissues and fresh tissue blocks from 49 patients with bladder cancer was detected by immunohistochemistry, real-time polymerase chain reaction (PCR) and Western blotting. Results The expression of Twist protein was detected in 14 cases (26.66% ) of bladder cancer. Twist positive rate was associated with the pathological parameters. The expression of Twist was closely associated with epithelial-mesenchymal transition and infiltration and metastasis. Conclusion Twist has an effect on epithelial-mesenchymal transition in bladder cancer. Twist may play an important role in the infiltration and metastasis of bladder cancer.
