益气活血法对脑缺血再灌注损伤后神经可塑性影响的实验研究

摘    要:目的探讨益气活血法对局灶性脑缺血再灌注损伤大鼠缺血半暗带神经可塑性的影响及作用机制。方法120只SD大鼠分为假模型组、手术组、益气活血法组、活血法组、益气法组。每组再分为缺血再灌注3、7、14d三个亚组。以线栓法阻塞大鼠左侧大脑中动脉,复制局灶性脑缺血再灌注模型,缺血2h再灌注3、7、14d,用免疫组织化学染色SABC法分别检测缺血半暗带突触素SYP、生长相关蛋白GAP-43的表达。结果缺血再灌注3d,益气组、活血组SYP的表达与模型组比较,差异有统计学意义(P〈0.05),益气活血组、活血组的GAP-43表达,与模型组比较,差异有统计学意义(P〈0.05);缺血再灌注7、14d,活血组和益气活血组SYP的表达与模型组比较,差异有统计学意义(P〈0.05),益气活血组GAP-43表达与模型组比较,差异有统计学意义(P〈0.05)。结论益气活血法能促进SYP、GAP-43表达,有利于脑缺血后神经细胞轴突的再生以及突触的重塑。

Experimental Research of the Impacts on Neural Plasticity after Cerebral Ischemic Reperfusion Injury Treated with the Therapy for Benefiting Qi and Activating Blood Circulation

Abstract:Objective To explore the impacts and mechanism on neural plasticity in ischemic penumbra for the rats with focal cerebral ischemic reperfusion injury treated with the therapy for benefiting qi and activating blood circulation. Methods 120 SD rats were divided into a sham - model group, an operation group, a benefiting qi and activating blood circulation method group( A group), an activating blood circulation method group( B group)and a benefiting qi method group( C group). Each group was subdivided into three subgroups corresponding to 3,7 and 14 d of ischemic reperfusion. The suture method was adopted to occlude the left middle cerebral artery(MCA) in the rats and duplicate the models of focal cerebral ischemic reperfusion. With ischemia for 2 h, the reperfusion was done for 3,7 and 14 d separately. The immunohistochemical staining SABC method was used to detect the expressions of synaptophysin(SYN) of ischemic penumbra and the growth - associated protein - 43 ( GAP - 43 ). Results Statistically, after ischemic reperfusion for 3 days, SYP expression in B and C groups was significantly different as compared with the model group ( P 〈 0.05 ) ;GAP -43 expression in A and B group was significantly different as compared with the model group (P 〈 O. 05 ) ;in 7 and 14 days of ischemic reperfusion, SYP expression in A and B groups was significantly different as compared with the model group (P 〈 0.05 ) ;GAP -43 expression in A group was significantly different as compared with the model group (P 〈 O. 05). Conclusion The therapy for benefiting qi and activating blood circulation increases SYP and GAP -43 expressions and contributes to the neuronal axon regeneration and synaptic remodeling after cerebral ischemia.
