热休克蛋白47在糖尿病大鼠皮肤创口愈合过程中的表达

摘    要:目的:使用糖尿病大鼠模型探讨热休克蛋白47(47kDa heat shock protein,HSP47)在糖尿病大鼠皮肤创口愈合过程中的表达水平。方法:在距正常大鼠和糖尿病大鼠头部1cm的背部做一直径1cm的圆形全层皮肤创口,糖尿病大鼠分为不注射组、注射空质粒组和注射重组质粒组,在创口愈合的第3、5、7天分别取对照组和实验组大鼠的创周皮肤,应用免疫组织化学和Western免疫印记法(Western blotting)检测HSP47在各组大鼠皮肤创口愈合过程中的表达水平。结果:正常大鼠皮肤创周组织中HSP47的表达,在愈合过程中呈明显增高的趋势。同正常大鼠相比,糖尿病大鼠皮肤创口愈合缓慢,皮肤创口组织中HSP47的表达明显减少。注射重组质粒pTracer-CMV-HSP47的糖尿病大鼠皮肤创周组织中HSP47的表达增强。结论:HSP47在糖尿病大鼠皮肤创口组织中的低表达与糖尿病大鼠皮肤创口愈合缓慢密切相关。通过体外构建HSP47的重组质粒再注入糖尿病大鼠皮肤创周组织中的方法,可以增强HSP47在体内的表达。

Abstract:Objective: To investigate the expression of 47kDa heat shock protein(HSP47) during wounds healing in diabetic rat skin. Methods: A circular full-thickness skin wound(1cm diameter) was created on the back skin of normal rats and diabetic rats, about 1 cm from the skull base.The diabetic rats were divided into three groups:noninjected group, injected pTracer-CMV group and injected pTracer-CMV-HSP47 group. Periwound skin tissues were examined at 3rd, 5th and 7th day. Immunohistochemical and Western immunoblotting were adopted to detect the expression of HSP47 in the tissues during healing of rat skin wounds. Results: Expressions of HSP47 remarkedly increased during healing process in normal rats. Expression of HSP47 during healing in diabetic rats wounds were much weaker. Being injected pTracer-CMV-HSP47, expression of HSP47 in the periwound tissues of diabetic rat increased. Conclusion: After being injected with pTracer-CMV-HSP47, expression of HSP47 increased in diabetic rat wound.
