摘    要:目的:研究辣椒碱对Hela(HPV+)和C-33A(HPV-)细胞生长增殖和凋亡的影响,探讨辣椒碱对宫颈癌细胞的抑制作用与相关机制。方法:选取HPV(+)的Hela和HPV(-)的C-33A宫颈癌细胞株为研究对象。CCK-8法检测细胞活性,流式细胞术检测辣椒碱对细胞生长的抑制作用、细胞周期和对细胞凋亡的影响;q PCR和Western blot法检测细胞凋亡相关基因m RNA和蛋白表达。结果:辣椒碱对宫颈癌Hela和C-33A细胞增殖抑制呈浓度和时间依赖性;辣椒碱能显著促进两种细胞凋亡,对Hela细胞的促凋亡效果更明显。辣椒碱处理后,两种细胞的细胞色素C(cytchrome C)和caspase3表达量显著增加,Bax表达量增加,Bcl-2表达量降低。结论:辣椒碱可显著抑制宫颈癌Hela和C-33A细胞增殖和促进细胞的凋亡,其机理可能是通过诱导细胞内活性氧类物质(ROS)增加、细胞色素C和Caspase3表达量增加等途径,最终诱导细胞凋亡。

Abstract:Objective:To observe the capsaicin on Hela (HPV+) and C-33A cell growth inhibition and influence on cell cycle and cell apoptosis.To explore the effects of capsaicin on cervical cancer cell inhibitory effect and the related mechanism.Methods:Two kinds of cervical cancer cell lines,HPV positive Hela and HPV negative C-33A cells,were chosen.Cell activity were examined by CCK-8 method,the inhibition of cell growth,cell cycles and cell apoptosis after capsaicin treatment were examined by flow cytometry.Cell apoptosis related gene mRNA and protein expression were detected by qPCR and Western blot.Results:The inhibitions of growth in Hela and C-33A cells after capsaicin treatment was getting severer along with the capaicin concentration increase and time prolongation.The capsaicin could promote the apoptosis of both kinds of cancer cells,and even more apoptosis of Hela cells.The expressions of cytochrom C,Caspase3 and bax were significantly increased,and the expression of Bcl-2 was significantly decreased.Conclusions:Capsaicin could effectively inhibit the growth and promote the apoptosis of Hela and C-33A.The anti-tumor effects of capsaicin might be achieved by cytchrome C pathway.Specifically,capsaicin may induce ROS increase in Hela and C-33A cell,then cytchrome C relive from mitochondria and trigere Caspase 3 apoptosis pathway in cell.
