不明原因复发性流产患者绒毛组织中miR-210表达及其与血管新生因子的关系

摘    要:目的:探讨不明原因复发性流产(URSA)患者绒毛组织中miR-210表达及其与血管新生因子的关系。方法:选取2014年3月至2016年4月在潍坊市益都中心医院产科行清宫术的URSA患者53例,同期选取正常早孕而终止妊娠的40例患者为对照组。实时荧光定量PCR法检测绒毛组织中miR-210、HIF-1α和VEGF表达,Western blot法检测绒毛组织中HIF-1α和VEGF蛋白表达,免疫组化法检测绒毛组织中MVD。结果:URSA患者绒毛组织中miR-210 mRNA相对表达量均低于对照组,且流产3次者的miR-210 mRNA相对表达量均低于流产≤3次的患者,差异均有统计学意义(P0.05)。URSA患者绒毛组织中HIF-1α和VEGF mRNA和蛋白相对表达量均低于对照组,且URSA患者中流产3次绒毛组织中HIF-1α和VEGF mRNA和蛋白相对表达量均低于流产≤3次的患者,差异均有统计学意义(P0.05)。URSA患者绒毛组织中MVD数(26.8±4.4)个,显著低于对照组的(37.2±5.1)个,差异有统计学意义(t=15.394,P0.001)。Pearson相关分析显示,URSA患者绒毛组织中miR-210与HIF-1α和VEGF表达均呈正相关(r=0.418和0.507,P0.05),与绒毛组织中MVD数呈正相关(r=0.472,P0.05)。结论:miR-210在URSA患者绒毛组织中呈低表达,且与流产次数有关,可能通过与HIF-1α和VEGF相互作用而减少MVD数量,导致URSA发生。

miR-210 expression in villus tissue of patients with unexplained recurrent spontaneous abortion and its relationship with angiogenesis factors
Abstract:Objective:To investigate the miR-210 expression in villus tissue of patients with unexplained recurrent spontaneous abortion (URSA) and its relationship with angiogenesis factors.Methods:53 cases of patients with URSA were selected from Mar.2014 to Apr.2016,40 cases of normal early pregnant women receiving artificial abortion were selected as control group.The expressions of miR-210,HIF-1α and VEGF in villus tissue were detected by using real-time fluorescence quantitative PCR.The expressions of HIF-1α protein and VEGF protein were detected by using Western blot.The micro lymphatic vessel density(MVD) in villus tissue was measured by immunohistochemical staining.Results:The relative expression levels of miR-210,HIF-1α mRNA and VEGF mRNA in villus tissue in URSA patients were lower than those of control group,and the relative expression levels of miR-210,HIF-1α mRNA and VEGF mRNA in villus tissue in URSA patients with abortion times (>3) times were lower than URSA patients with abortion times (≤3),the differences were statistically significant (P<0.05).The relative expression levels of HIF-1α protein and VEGF protein in villus tissue in URSA patients were lower than control group,and the relative expression levels of HIF-1α protein and VEGF protein in URSA patients with abortion times (>3) were URSA patients with abortion times (≤3),the differences were statistically significant (P<0.05).The MVD value in URSA patients was (26.8±4.4),which was significantly lower than(37.2±5.1) in control group,the difference was statistically significant(t=15.394,P<0.001).Pearson correlation analysis showed that the relative expression level of miR-210 in villus tissue in URSA patients was positively correlated with the relative expression level of HIF-1α mRNA,the relative expression level of VEGF mRNA,and MVD value (r=0.418,0.507 and 0.472,P<0.05).Conclusion:The expression of miR-210 was low in villi tissues in patients with URSA,and was related with the number of abortion.It might through interaction with HIF-1α and VEGF and MVD to reduce the number of MVD,and lead URSA occur.
