复黄片总鞣质、总黄酮及槐角苷的含量测定

目的建立复黄片中总鞣质、总黄酮及槐角苷的含量测定方法。方法以没食子酸和槐角苷为对照品,UV法测定复黄片中总鞣质及总黄酮的含量,检测波长分别为760和260nm。HPLC法测定复黄片中槐角苷的含量,采用Agilent TC-C18(2)色谱柱(250mm×4.6mm,5μm);柱温为30℃;流动相为乙腈-磷酸缓冲盐,采用梯度洗脱;流速为1.0mL·min-1;检测波长为254nm。结果复黄片中总鞣质、总黄酮及槐角苷的线性范围分别为1.04~5.20(r=0.999 5),4.15~9.34(r=0.999 5)和21.34~74.69μg·mL~(-1)(r=0.999 4),加样回收率分别为99.62%,100.20%和100.70%,平均含量分别为23.81,133.02和9.99mg·片~(-1)。结论该方法简便易行,结果准确、可靠,可用于控制复黄片的质量。

Determination of total tannins,total flavonoids and sophoricoside in Fuhuang Tablets

Objective To establish methods for the determination of total tanins ,total flavonoids and sophoricoside in Fuhuang Tab-lets .Methods The contents of total tanins and total flavonoids in Fuhuang Tablets were determined by UV with gallic acid and so-phoricoside as reference .The detection wavelengths were 760 and 260 nm .The separation of sophoricoside was performed on an Agilent TC-C18 (2) column (250 mm × 4 .6 mm ,5 μm) at 30 ℃ with gradient elution using phosphate buffer and acetonitrile as mobile phase at a flow rate of 1 .0 mL · min-1 .The detection wavelength was 254 nm .Results Linear ranges of total tanins ,total flavonoids and sophoricoside were 1 .04-5 .20 (r=0 .9995) ,4 .15-9 .34 (r=0 .9995) and 21 .34-74 .69 μg · mL -1 (r=0 .9994) , respectively .The average recoveries of accuracy tests were 99 .62% ,100 .20% and 100 .70% ,respectively .Contents of total tan-nins ,total flavonoids and sophoricoside in Fuhuang Tablet were 23 .81 ,133 .02 and 9 .99 mg · tablet-1 ,respectively .Conclusion The methods are simple and accurate .It can be used for the quality control of Fuhuang Tablets .
