摘    要:目的建立用HPLC法测定不同方法(熏干、新鲜和阴干)处理冬虫夏草样品中核苷类成分(腺苷、尿苷、肌苷、鸟苷和胸苷)含量的方法。方法样品用纯化水超声提取,流动相为甲醇-水,梯度洗脱,柱温:30℃,流速:1.0mL·min-1。结果虫草中含有的5种核苷标准曲线的相关系数均在0.999 0以上,精密度检测RSD值为0.31%~1.95%(n=5),重复性检测RSD值为0.50%~2.56%(n=5),回收率在99.46%~100.11%之间。结论该方法灵敏、简便、快速、准确、重复性好,能有效测定虫草中核苷类成分的含量,可为评价虫草的质量提供依据。

Abstract:Objective To determine the content of nucleosides (adenosine, uridine, inosine, guanosine and thymidine) in Cordyceps sinensis samples processed using different methods (smoked dry, fresh, dried) by HPLC.Methods The samples were extracted with purified water.The mobile phase was methanol-water, the column temperature was 30 ℃ and the flow rate was 1.0 mL·min-1.Results The calibration curves for the 5 nucleosides in Cordyceps sinensis displayed a good linear relationship with correlation coefficients above 0.999 0.The precision relative standard deviations (RSD) and the repeatability RSD of 5 standards were 0.31%-1.95%(n=5), 0.50%-2.56%(n=5), respectively.The recovery rate was in the range of 99.46%-100.11%.Conclusion This method is sensitive, simple, rapid, accurate and with good reproducibility so it can be used for the determination of the nucleosides in Cordyceps sinensis and provided an experimental basis for evaluating different Cordyceps sinensis more comprehensively.
