| 摘 要： | 目的:通过检测妊娠期糖尿病(GDM)患者胎盘中转化生长因子-β2(TGF-β2)及其受体TGFβRⅡmRNA和蛋白的表达,探讨两者在GDM发生中的作用。方法:实时定量PCR、Western blot和免疫组化方法检测30例GDM产妇和30例正常妊娠产妇胎盘组织中TGF-β2及TGFβRⅡ的表达情况。结果:GDM组胎盘组织中TGF-β2和TGFβRⅡmRNA和蛋白表达水平均显著高于正常妊娠组(P0.05);GDM组胎盘中TGF-β2和TGFβRⅡ阳性表达率均高于正常妊娠组,差异均有统计学意义(P0.05)。结论:GDM胎盘组织中TGF-β2及TGFβRⅡ升高可能与GDM发生有关,两者可能成为新的诊断标志物和治疗GDM的靶点。

| Abstract: | Objective:To investigate the expression and function of transforming growth factor beta-2 ( TGF-β2 ) and transforming growth factor beta receptor 2 ( TGFβRⅡ) in gesta-tional diabetes mellitus ( GDM) . Methods:The placentas from 30 GDM patients and 30 normal pregnant women were collected. The expressions of TGF-β2 and TGFβRⅡwere detected by re-al-time PCR,Western blot and immunohistochemistry. Results:The mRNA and protein expres-sions of TGFβ2 and TGFβRⅡ were significantly higher in GDM group than those in control group ( P<0 . 05 ) . The positive expression levels of TGFβ2 and TGFβRⅡ were significantly higher in GDM than those in control group(P<0. 05). Conclusions:The up-regulations of TGF-β2 and TGFβRⅡ might be associated with the etiology of GDM,TGF-β2 and TGFβRⅡmight be targets of diagnosis and treatment in GDM. |
