草苁蓉提取物在大鼠肝脏化学致癌初期对血清一氧化氮含量和一氧化氮合酶活性的影响

摘    要:[目的 ]探讨草苁蓉乙醇提取物再经水和二氯甲烷萃取的水层部分在大鼠肝脏化学致癌初期对一氧化氮含量和一氧化氮合酶活性的影响 ,从而进一步探讨其抗致癌作用的可能机理 .[方法 ]按略改良的Solt Farber氏方法制作大鼠肝脏癌前病变模型 ,用二氯化甲烷萃取水层部分 ,饲养大鼠 6周后处死 ,取血液 ,以比色法测定血清一氧化氮含量和一氧化氮合酶活性 .[结果 ]草苁蓉给药组血清一氧化氮含量低于癌前病变模型组 ;草苁蓉给药组血清一氧化氮合酶活性低于癌前病变模型组 .[结论 ]草苁蓉乙醇提取物再经进一步分离的水层成分可降低一氧化氮含量、一氧化氮合酶水平 ,而这可能是其抗致癌作用机理之一

Abstract:OBJECTIVE To study the effects of water layer separated from alcohol extract of Boschniakia rossica on content of nitric oxide and activity of nitric oxide synthase in early stage of rat chemical hepatocarcinogenesis and some of mechanism of its anti-carcinogenesis. METHODS Hepatic precancerous lesions in rats were induced by the modified solt-farber protocol and the rats were killed after feeding with the water extracted from dichloromethane for six weeks. The serum content of nitric oxide and activity of nitric oxide synthase were measured by colorimetric method. RESULTS The serum content of nitric oxide in Boschniakia rossica group fed with Boschniakia rossica was lower than that in the nitric oxide synthase odel group of early stage of chemical hepatocarcinogenesis and the activity of in Boschniakia rossica group was lower than that the model group. CONCLUSION The further seperated water layer ingredient of alcohol extract of Boschniakia rossica could reduce the content of nitric oxide and activity of nitric oxide synthase, and it may be one of the anti-carcinogenic mechanisms.
