培哚普利对胰岛素抵抗的影响

摘    要:目的 :研究培哚普利对胰岛素抵抗高血压大鼠胰岛素抵抗 (IR)的影响。方法 :选择雄性SD大鼠 30只分成 2组 ,对照组饲以基础饲料 ,培哚普利组饲以高糖高盐饲料 ,4wk后用培哚普利 0 .8mg·kg- 1·d- 1灌胃 4wk ,观察 2组实验前、4wk、8wk时收缩压 (SBP)、空腹血糖 (FBG )、空腹血胰岛素 (FINS)、胰岛素敏感性指数 (ISI)及红细胞膜Na ,K ATP酶活性测定的变化。结果 :培哚普利组 4wk时SBP升高 ,上升值为 (4 .4±s 1.6 )kPa ,FBG上升值 (0 .4± 0 .3)mmol·L- 1,FINS升高 (12 .7±2 .1) μU·L- 1,ISI降低 ,差值 1.30± 0 .2 0 ,红细胞膜Na ,K ATP酶活性降低 (12± 6 )nmoL·pi·mg- 1·pro·h- 1,P <0 .0 1。 8wk与 4wk时比较分别为 ,SBP[(13.2± 1.2 )kPavs (17.0± 2 .0 )kPa](P <0 .0 1) ,FBG分别为 [(2 .9± 0 .7)mmol·L- 1vs (3.8± 0 .3)mmol·L- 1](P <0 .0 1) ,FINS分别为 [(8.0± 1.5 ) μU·L- 1vs (18± 3) μU·L- 1](P <0 .0 1) ,ISI分别为 [(- 3.2 0± 0 .2 0 )vs (- 4.2 0± 0 .2 0 ) ](P <0 .0 1) ,红细胞膜Na ,K ATP酶活性分别为[(36± 9)nmoL·pi·mg- 1·pro·h- 1vs (2 6± 6 )nmoL·pi·mg- 1·pro·h- 1](P <0 .0 1)。结论 :培哚普利能改善高糖高盐饲养大鼠的IR。

Abstract:AIM: To investigate the influence of perindopril on insulin resistance to hypertensive rat model (high glucose high salt fed rats(GSFR)). METHODS: Male Spraque-Darley 30 rats were divided into two groups (control group and perindopril group).The perindopril group was fed on high glucose and high salt diet(50% glucose 8% salt by weight) for 4 wk and treated with perindopril 0.8 mg·kg -1·d -1 for the later 4 wk. Another rats were fed with standard food for 8 wk. The changes of systolic blood pressure (SBP),fasting blood glucose (FBG),fasting blood insulin (FINS),insulin sensitivity index (ISI), and Na,K-ATPase activity of erythrocyte membrane of two groups before experiment, 4 wk and 8 wk were observed. RESULTS: In perindopril group of 4 wk, the SBP, FBG and FINS increased, SBP (4.4±s 1.6) kPa,FBG (0.4±0.3) mmol·L -1,and FINS(12.7±2.1) μU·L -1 , ISI decreased(1.30±0.20), and Na,K-ATPase activity decreased(12±6) nmoL·pi·mg -1pro·h -1, (P< 0.01). This indicated that hypertension and insulin resistance wasproduced in rars. The comparison of following times between 8 wk and 4 wk:SBP and FINS decreased, SBP((13.2±1.2)kPa vs (17.0±2.0) kPa) (P<0.01), FBG decreased ((2.9± 0.7) mmol·L -1 vs (3.8± 0.3) mmol·L -1) (P< 0.01), FINS ((8.0± 1.5) μU·L -1 vs (18±3) μU·L -1) (P< 0.01),ISI increased ((-3.20± 0.20) vs (-4.20± 0.20)) (P< 0.01)and Na,K-ATPase activity increased((26±6) nmoL·pi·mg -1pro·h -1 vs (36±9) nmoL·pi·mg -1pro·h -1) (P< 0.01). CONCLUSION: Perindopril can improve insulin resistance (IR) to the GSFR.
