口腔鳞状细胞癌基质金属蛋白酶-9表达及其与血管生成的关系

摘    要:目的]探讨口腔鳞状细胞癌(OSCC)发生,发展过程中基质金属蛋白酶-9(MMP-9)对微血管生成的意义。方法]采用疫组化SP法对65例OSCC、40例非典型增生、22例正常口腔黏膜进行MMP-9、CD34免疫组化染色。检测OSCC、非典型增生、正常口腔黏膜中MMP-9的表达和微血管密度(MVD).并分析MMP-9的表达与MVD之间,以及它们与OSCC临床病理特征之间的关系。结果] OSCC组中MMP-9的表达与MVD值均高于非典型增生组,在非典型增生组中均高于正常口腔黏膜组:OSCC组、非典型增生组和正常口腔黏膜组中MMP-9表达与MVD有正相关趋势;MMP-9的表达与肿瘤大小、淋巴结转移密切相类.MVD与淋巴结转移密切相炎。结论] MMP-9可能是OSCC的重要促血管生成因于之一。

Correlation between Expression of Matrix Metalloproteinase-9 and Angiogenesis in Oral Squamous Cell Carcinoma

Abstract:Purpose] To study the significance of matrix metalloproteinases-9 (MMP-9) on micro angiogenesis in carcinogenesis and progression of oral squamous cell carcinoma (OSCC). Methods] MMP-9 and CD34 were detected by immunohistochemistry SP method in 22 cases of normal oral mucosa, 40 cases of atypical hyperlasia and 65 cases with OSCC. The expression of MMP-9 and microvessel density (MVD) were detected in the three groups and the relationship between expression of MMP-9 and MVD and its relationship with OSCC clinicpathological characteristics were reviewed.Results] The expression of MMP-9 and the level of MVD in OSCC were significantly higher than that in atypical hyperlasia. The expression of MMP-9 and the level of MVD in the group of atypical hyperlasia were significantly higher than that in normal oral mucosa. The expression of MMP-9 were closely related with MVD in the tumor group, atypical hyperlasia group and normal oral mucosa group. MMP-9 expression in OSCC was significantly associated with lymph node metastasis and tumor size. MVD in OSCC was significantly associated with lymph node metastasis. Conclusion] MMP-9 may be one of important factors in angiogenesis of OSCC.
