摘    要:目的 观察四种Notch配体的功能活性.探讨其在Notch信号传导机制中的作用。方法以Notch1—CHO和Notch2—CHO作为宿主细胞,瞬时转染报告质粒TP1,分别加入转染有不同Notch配体Delta1、Jagged1、Jagged2、Delta4的CHO细胞及未转染CHO细胞,使Notch分子与其配体充分结合,加入发光底物PGL100,用Lumat测定发光情况,由此观察并比较各配体对上述两种宿主细胞的信号功能活性水平。结果 各配体与Notch1结合后均能引起荧光素酶活性增高,尤其以Jagged2—CHO、Delta4—CHO1—4、Delta4—CHO1—5为著,而Deltal—CHO、Jagged1—CHO引起的活性增加程度较低,尚不能确定其信号功能活性;各配体与Notch2结合后都有较高的信号功能活性,其反应强于与Notch1的作用。结论 4种配体与Notch2作用后均能使荧光素酶活性增加多倍,故有较强的信号功能活性。

Abstract:Objective To find out the function of four Notch ligands in Notch signal transduction mechanism.Methods Notch1-CHO and Notch2-CHO was used as the host cells,transiently transfected with reported plasmid TP1,and employed Luciferase analysis to test the interaction between Notch and Notch ligands,then observed and compared the activity of four Notch ligands.Results All of the four Notch ligands bindings to Notch1 could induce TP1 activity,especially Jagged2-CHO,Delta4-CHO1-4,Delta4-CHO1-5,Delta4,but Delta1-CHO,Jagged1-CHO increased the activity lowerly and it can noten sure their signal activity.Conclusion For Notch2,the TP1 activity enhanced by the four ligands in these cells was much higher than that of CHO.
