摘    要:血管内皮细胞具有纤溶功能,其表现之一为内皮细胞表面的特异性受体与纤溶系统的启动因子——组织型纤溶酶原激活物(tPA)结合,结合后tPA活性明显增高且免受其抑制物(PAI)的灭活,从而使内皮细胞纤溶活性升高。本实验用联胺作用于内皮细胞,诱发内皮细胞过氧化损伤,用放射性核素测定及放射自显影方法观察内皮细胞损伤前后与tPA结合力的改变。结果表明:内皮细胞过氧化损伤后与tPA结合力明显下降。提示内皮细胞损伤后纤溶活性降低的可能机制与二者结合力下降有关。

Abstract:Tissue plasminogen activator(t PA) have been proved playing an important role in the fibrinolytic system. After t PA bound to endothelial cell (EC), the apparent catalytic efficiency of t PA would be further increased, and the binding would also prevent t PA from the inhibition of its inhibitor(PAI). In this paper, Diamine was used to cause the injury by lipoperoxidation to cultured human umbilical vein endothelial cells.Radioassay and autoradiography were applied to study the effect of the injury on the binding ability of t PA to EC. The outcomes indicated that the binding of t PA to EC decreased obviously after the injury to EC by lipoperoxidation, suggested that the reduction of the fibrinolytic activities induced by EC related to the decrease of the binding ability of t PA to EC due to the injury.
