摘    要:目的:检测转基因水牛人FSHβcDNA序列基因。方法:分离提取转基因水牛外周血DNA,PCR扩增,T-A克隆测序,DNAclub软件分析。结果:水牛基因组整合人FSHβcDNA序列并发生一位点缺失C44(无意义链)或G347(有意义链),突变位于人FSHβ-Seatbelt结构域。结论:转基因水牛整合人FSHβcDNA序列发生点缺失,导致阅读框架发生移码,其功能有待进一步研究。

Abstract:Objective To identify human FSHβ gene in the transgenic buffalo. Method The DNA was isolated from the blood tissue of transgenic buffalo. The PCR product for human FSHβ cDNA was cloned into the pGEM-T easy vector nd was detected by DNA sequencing. Results The detected sequences from buffalo DNA was human FSHβ cDNA;which there was only one deletion in the site of C44 (non-sense strand) or G347(sense-strand),The deletion was located in the domain of human FSHβ-seat belt by DNA club software analysis. Conclusion The cDNA of human FSHβ is incorporated into the buffalo genome which deletion remains further research.
