软脉宁对高脂血清诱导内皮细胞损伤的影响

摘    要:以兔高脂血清培养内皮细胞(EC)造成离体动脉粥样硬化(AS)模型,应用相差显微镜、电镜、脂蛋白(a)Lp(a)]、组织型纤溶酶原激活物(t-PA)及其抑制物(PAI)的测定,观察软脉宁对脂质过氧化损伤的EC形态、结构和纤溶功能的保护作用.结果表明:软脉宁可使由脂质过氧化作用造成的EC细胞收缩及生物膜系统损伤的程度减轻,并使LP(a)浓度显著降低、t-PA活性显著增高、PAI活性显著降低,且LP(a)与PAI呈正相关.提示:软脉宁通过减轻EC脂质过氧化损伤程度,降低LP(a)浓度,维护了纤溶功能的平衡,抑制血栓的生成,从而阻止了AS的形成和发展.

Abstract:The model of atherosclerosis in vitro was established by culturing newborn endothelial cells (EC) of umbilical veins with rabbit high lipid serum, and the protective effects of Ruanmaining Decoction (RD) against the injury induced by lipid peroxidation on EC form, structure and fibrinolytic function were observed by the phase contrast microscopy, electron microscopy, and by the detection of lipoprotein (a) (LP(a)), tissue plasminogen activator (t PA) and plasminogen activator inhibitor (PAI). The results showed that RD could reduce EC shrinkage and the degree of the injury of EC biological membrane system. The results also showed that RD could markedly decrease the level of LP(a), increase the activity of t PA, and reduce the activity of PAI, with a positive correlation between LP(a) and PAI.
