食欲素对小鼠胃肠道功能及胃肠激素分泌的影响

摘    要:目的探讨食欲素对外周胃肠道消化功能的影响。方法将小鼠随机分为溶剂对照组(A组),食欲素低、中、高剂量组[25μg/kg(B1组)、50μg/kg(B2组)、75μg/kg(B3组)],B3组+10 mg/kg食欲素1型受体拮抗剂处理组(C组),B3组+10 mg/kg食欲素2型受体拮抗剂处理组(D组)。观察各组小鼠每日摄食量,检测其胃液的分泌量和胃蛋白酶的活性;计算小肠炭末推进率,检测小肠的运动功能。结果与A组比较,B组小鼠摄食量、胃液分泌量及小肠运动功能显著增加(P<0.05),且具有浓度依赖性;C组给予食欲素1型受体拮抗剂后食欲素促进摄食、小肠运动功能和胃液分泌的作用消失(P>0.05);D组给予食欲素2型受体拮抗剂后食欲素仍能促进摄食、增加小肠运动功能和胃液分泌,提高胃蛋白酶活性(P<0.05)。结论食欲素能通过激活1型受体,增强外周胃肠道消化功能。

Abstract:Objective To investigate the effect of orexin on gastrointestinal digestive function.Methods Mice were randomly divided into the vehicle control group(group A),orexin treatment group[25μg/kg(group B1),50μg/kg(group B2),and 75μg/kg(group B3)],orexin(75μg/kg)+10 mg/kg orexin 1 receptor antagonist group(group C),and orexin(75μg/kg)+10 mg/kg orexin 2 receptor antagonist group(group D).The daily food intake of mice was observed,and the secretion of gastric juice and the activity of pepsinate were detected.The movement function of small intestine was measured by calculating the propulsive rate of carbon powder in small intestine.Results Compared with group A,the animal's food intake,gastric juice secretion and small intestinal motility in group B increased significantly(P<0.05)and were concentration-dependent.The effect of orexin in promoting food intake,small intestinal motility and gastric juice secretion disappeared after the administration of orexin-1 receptor antagonist in group C(P>0.05).While orexin still promoted food intake,increased small intestinal motion and secretion of gastric juice and promoted the activity of pepsin in group D(P<0.05).Conclusion Orexin could promote the gastrointestinal digestive function via stimulating orexin 1 receptor.
