半枝莲提取物通过PI3K-Akt信号通路抑制胰腺癌模型大鼠肿瘤生长的机制研究

摘    要:目的:基于PI3K-Akt信号通路探究半枝莲提取物对胰腺癌模型大鼠的干预效果。方法:选取40只SD健康雄性大鼠,10只为正常组,其余30只建立胰腺癌模型,分为模型组、低浓度半枝莲组、高浓度半枝莲组。检测各组大鼠肿瘤体积、重量、T淋巴细胞亚群水平、细胞凋亡及Wnt3α、β-catenin、PI3K、Akt、mTOR表达。结果:高浓度半枝莲组大鼠肿瘤体积、重量低于低浓度半枝莲组、模型组(均P<0.05)。高浓度半枝莲组大鼠CD8+水平低于低浓度半枝莲组、模型组,CD4+、CD3+水平高于低浓度半枝莲组、模型组(均P<0.05)。高浓度半枝莲组大鼠Wnt3α、β-catenin、PI3K、Akt、mTOR表达低于低浓度半枝莲组、模型组(均P<0.05)。结论:半枝莲提取物可抑制胰腺癌模型大鼠肿瘤组织,调控胰腺癌大鼠T淋巴细胞亚群水平,调控Wnt/β-catenin、PI3K-Akt信号通路相关蛋白表达,促进胰腺癌细胞自噬、凋亡。

Abstract:Objective:The intervention effect of scutellaria barbata extract on pancreatic cancer model rats was explored based on PI3K-Akt signaling pathway.Methods:40 SD healthy male rats were selected,10 cases normal groups,30 cases established pancreatic cancer models,which were divided into model group,low concentration group,high concentration group.The levels of tumor volume,weight,T lymphocyte subsets,apoptosis,Wnt3α,β-catenin,PI3K,Akt,mTOR were compared between the four groups.Results:The tumor volume and weight of high concentration scutellaria barbata group were lower than those of the low concentration scutellaria barbata group and the model group(all P<0.05).The level of CD8+ in high concentration scutellaria barbata group was lower than that in the low concentration scutellaria barbata group and the model group,the level of CD4+ and CD3+ in the high concentration scutellaria barbata group were higher than those in the low concentration scutellaria barbata group and the model group(all P<0.05).The expression of Wnt3α,β-catenin,PI3K,Akt and mTOR in the high concentration scutellaria chinensis groups were lower than those in the low concentration scutellaria chinensis group and the model group(all P<0.05).Conclusion:Scutellaria barbata extract was able to inhibit tumor tissue of the pancreatic cancer model rats,and regulate the level of T lymphocyte subsets in pancreatic cancer rats,regulate Wnt/β-catenin pathway and PI3K-Akt signaling pathway protein expression,promote apoptosis of pancreatic cancer cells.
