摘    要:目的探讨ABCG2在肝癌细胞的耐药及转移侵袭过程中的作用。方法 MTT方法检测siRNA干扰前后三种肝癌细胞MHCC97-L、MHCC97-H、SMMC-7721对多柔比星的药物敏感性;划痕与Transwell实验证实这几种肝癌细胞在siRNA干扰前后的转移、侵袭能力;western-blot、PCR实验证实siRNA转染效率。结果 SMMC-7721、MHCC97-L、MHCC97-H细胞内ABCG2水平呈逐渐上升趋势,对多柔比星的耐受程度也同样逐步上升,同时侵袭能力逐渐增强;siRNA干扰后,MHCC97-L、MHCC97-H两株细胞中ABCG2水平明显降低,对多柔比星敏感性明显增强,侵袭能力明显下降。结论 ABCG2不仅在肝癌细胞的耐药过程中起到重要作用,同时还影响肝癌细胞的转移侵袭能力,靶向干扰ABCG2水平增加了化疗药物敏感性并降低肝癌细胞的转移侵袭能力。

ABCG2 affects the chemoresistance and invasion ability in HCC after interfered by siRNA

Abstract:Objective To determine the relationship between the potential as determined by MTlr assay while the migration and invasion ability was measured by wound-healing assay and transwell assay, respectively. Results Of all the three HCC cell lines, SMMC-7721, MHCC97-L and MHCC97-H showed ascending ABCG2 expression, at the same time, their resistance to doxorubicin and invasion ability increased. After we interfered the ABCG2 level, the ABCG2 overexpression cell lines MHCC97-L and MHCC97-H be- came more sensitive to doxorubicin and their invasion ability weakened. Conclusions ABCG2 not only gives rise to the ehemoresis- tance in HCC but also affects its invasion ability. ABCG2 targeted therapy might increase the sensitivity of HCC chemotherapeutic drugs and reduce the migration and invasion ability.
