摘    要:目的 探讨重线人粒细胞集落刺激因子(rhu G-CSF)对K562细胞凋亡和其细胞毒药物敏感性的影响。方法 采用流式细胞术和MTT法分别测定细胞凋亡和细胞毒药物敏感性。结果 在低血清培养条件下,rhu G-CSF对K562细胞的凋亡有抑制作用,其凋亡百分率与rhu G-CSF浓度呈负线性样;rhu G-CSF可增加K562细胞对Ara-C的敏感性,而降低其对DNR的敏感性,在Ara-C和DNR联合

Abstract:Objective To investigate the effect of human recombinant granulocyte-clony stimulating factor (rhu G-CSF) on the apoptosis of K562 cell and its sensitivity to cytotoxic agents. Methods Cell apoptosis and sensitivity of cytotoxic agents were analyzed by flow cytometry and MTT assay respectively. Results At lower serum concentration, the rhu G-CSF had an inhibitory effect on the apoptosis of K562 cell in a negative dose dependent manner. rhu G-CSF enhanced the sensitivity of K562 cell to Ara-C while decreased its sensitivity to daunorubicin. rhu G-CSF enhanced the combinant cytotoxicity of Ara-C and daunorubicin to K562 cells. Conclusion rhu G-CSF can enhance the combined cytotoxicity of Ara-C and daunorubicin to K562 cells.
