Abstract:Objective To determine the effect of 2′,5,6′,7-tetrahydroxyflavanonol pretreatment on the release of cytokine(TNF-α and IL-6) and the expression of TNF-α mRNA in RAW264.7 cells after exposure to LPS.Methods RAW264.7 cells were pretreated with THF at the indicated dose(0,10,20,40,80,160 mg/L) and stimulated with LPS(100 μg/L).Meanwhile,ELISA and Real-Time PCR were used respectively to measure the release of cytokine(TNF-α and IL-6) and detect the expression of TNF-α mRNA in RAW264.7 cells.The cytotoxicity of THF on RAW264.7 was established using the MTT assay in order to remove the effect of THF pretreatment on cell viability.Results THF pretreatment at different concentrations significantly decreased the release of TNF-α and IL-6 in LPS-induced RAW264.7 cells in a dose-dependent manner.Furthermore,THF reduced the expression of TNF-α mRNA in LPS-induced RAW264.7 cells in a dose-dependent manner.MTT results showed that THF treatment below 320 mg/L concentration did not affect RAW264.7 cell viability.Conclusion THF can antagonize LPS stimulation on the effects of inflammation cells and is expected to be a candidate prodrug for anti-sepsis.
