脂肪肝肝组织脂质含量与肝功能关系的实验研究

目的探讨脂肪肝肝组织脂质含量与肝功能的关系。方法64只SPF级Wistar大鼠,随机分为5组,A组、B组、C组、D组、E组给予高脂饲料(84.4%标准饲料+10%猪油+0.5%胆固醇+0.1%胆盐+5%蛋黄粉)和白酒复合复制大鼠脂肪肝模型,B组、C组、D组、E组分别给予肝康Ⅳ号(GKⅣ)低、中、高剂量和东宝肝泰灌胃干预,设空白对照F组。第6周末处死动物取肝脏制备10%的肝匀浆查胆固醇(TC)、甘油三脂(TG),腹主动脉抽血检测丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)。结果B组、C组、D组、E组与A组比较,TC、TG含量明显下降(P<0.05～0.01),ALT、AST水平明显升高(P<0.05～0.01);在TC、TG方面,C组、D组与E组比较差异有统计学意义(P<0.05);在ALT、AST方面C组、D组与E组比较差异有统计学意义(P<0.01)。结论GKⅣ能有效降低肝脏组织脂质沉积,防止ALT、AST的升高;ALT、AST与TC、TG的沉积量有密切的关系。

The experimental study on relation between lipid levels of fatty liver and liver function

Objective To explore relation between lipid levels of fatty liver and liver function.Methods 64 SPF wistar rats were randomly divided into Group A,B,C,D,E and F.Group A,B,C,D and E were given high fat diet(84.4% standard food,10% lard,0.5% cholesterol,0.1% bile salt and 5% egg yolk powder)and wine to make fatty liver model of rats,group B,C,D and E were respectively given low,middle and high dose of Gankang Ⅳ(GKⅣ)and Dongbao Gantai gavaged intervention,and a control group F was set.After 6 weeks,rats were sacrificed and 10% liver homogenate was made from liver to test TC and TG,abdominal aortic blood was drawed to test ALT and AST.Results Compared with group A,TC and TG levels decreased significantly(P<0.05～0.01),ALT and AST levels were significantly higher(P<0.05～-0.01)in group B,C,D and E;Compared group C and D with E there was significant difference for TC and TG(P<0.05);For ALT and AST,compared group C and D with E there was a very significant difference(P<0.01).Conclusion GKⅣ can effectively reduce the liver lipid deposition;inhibit increasing of ALT and AST.From the indicators of ALT,AST,TC and TG,the deposition amount of TC and TG have a close relationship with ALT and AST.
