鞘内注射新斯的明和吗啡对大鼠脊髓背角c-fos表达的影响

摘    要:目的 研究鞘内注射新斯的明和吗啡对大鼠脊髓背角c-fos表达的影响。方法 在大鼠切口疼痛模型上结合鞘内置管技术,采用累积疼痛评分法评定大鼠疼痛行为,运用免疫组织化学法观察术前或术后鞘内注射新斯的明和吗啡对脊髓背角c-fos表达的影响。结果 手术组大鼠累积疼痛评分明显高于假手术组,术前或术后鞘内注射新斯的明及吗啡均可明显降低手术切口引起的累积疼痛评分升高,各用药组间无明显差别。手术组大鼠脊髓背角Fos-LI神经元明显多于假手术组(P<0.01)。术前或术后鞘内注射吗啡、新斯的明和吗啡分别可使切口疼痛引起的Fos-LI减少69%和48%,76%和55%,术前和术后两用药组间比较差异亦有统计学意义。新斯的明两用药组的Fos-LI表达均未见明显减少(P>0.05)。结论 鞘内注射新斯的明和吗啡可减少术后疼痛大鼠脊髓背角c-fos的表达,尤以术前用药为甚。

Abstract:Objective To study the effects of intrathecal neostigmine and morphine on c - fos expression in the dorsal horn of spinal cord in rats. Methods In a rat model of postoperative pain, cumulative pain scores were used to evaluate pain behaviors in combination with intrathecal injection. Immunohistochemistly technique was used to measure c - fos expressions in spinal cord of rats and the effects of pre - or post - operative intrathecal neostigmine, morphine and neostigmine with morphine. Results The cumulative pain scores of rats in operation group were significantly higher than those of sham - operation rats. Intrathecal neostigmine, morphine and neostigmine with morphine either before or after operation significantly decreased cumulative pain scores. There were no significant differences among drug- used groups, c- fos expression decreased by 69% and 48%, 76% and 55% when morphine, neostigmine with morphine were used before or after operation( P < 0.01) . Neostigmine did not significantly reduce c - fos expression( P > 0.05) . Conclusion Intrathecal morphine and neostigmine with morphine significantly decrease c- fos expression in the dorsal horn of spinal cord in rats. The effects are better when drug was given before operation.
