丹黄通脉方对大鼠颈动脉粥样硬化性狭窄模型炎性反应的影响

摘    要:目的研究丹黄通脉方(DHTMD)对大鼠颈动脉粥样硬化性狭窄模型炎性反应的影响。方法将大鼠随机分为四组,每组10只。三组采用高脂加高蛋氨酸复合饲料喂养4 W后行颈总动脉球囊拉伤术法制备大鼠颈动脉狭窄模型,空白组给予普通饲料喂养,其余三组给予高脂加高蛋氨酸复合饲料喂养,阳性药组给予辛伐他汀2.5 mg/kg,DHTMD组给予DHTMD 20 g/kg生药量,配制成10 mL/kg的药液,灌胃给药,一天两次。继续喂养8 W后,检测血脂四项、TNF-α、CRP、ICAM-1及Hcy的水平。结果与空白组比较,4W时其它三组大鼠的TC和Hcy明显升高(P<0.05);治疗8 W后与模型组比较,DHTMD组和阳性药组的Hcy、CRP、TNF-α、ICAM-1、TC、TG、LDL-C水平均有显著降低(P<0.05);与阳性药组比较,DHTMD组的Hcy、CRP、TNF-α、ICAM-1及LDL-C水平降低且有统计学意义(P<0.05)。结论 DHTMD具有调节血脂代谢和抗炎作用,能够抑制颈动脉粥样硬化的形成,防治缺血性脑血管疾病。

Abstract:Objective To observe the inlfluence of Dan Huang Tong Mai decoction(DHTMD) on the inflammatory reaction of the carotid atherosclerotic stenosis rats model.Methods The rats were randomly divided into four groups,10 in each group.Three groups were used to prepare the carotid atherosclerotic stenosis rats model after fed with compound feed of high-fat and high-methionine in 4 weeks,the rest of the group named control group was fed with normal food.After 4 weeks,three groups were still fed with compound feed,and on this basis,the positive medicine group was given Simvastatin 2.5 mg/kg,while the DHTMD group were given DHTMD 20 g/kg born dosage,made into 10 mL/kg solution,adiministration oral twice a day.8 weeks later,the blood fat four items,TNF-ɑ,CRP,ICAM-1and Hcy were determined.Results Compared with control group,the level of TC and Hcy in other three groups were obviously increased at fourth week(P<0.05);After 8 weeks,compared with model group,the level of Hcy,CRP,TNF-α,ICAM-1,TC,TG and LDL-C both in DHTMD and positive medicine group were significantly reduced(P<0.05);Compared with positive medicine group,the Hcy,CRP,TNF-α,ICAM-1 and LDL-C in DHTMD group were decrease(P<0.05) and had statistical difference.Conclusion DHTMD has the effect not only on regulation of the blood fat metabolism and inflammation,but also on inhibition the formation of carotid atherosclerosis and prevention of ischemic cerebral vascular disease.
