大鼠感染及无菌性炎症模型创口渗出液的酶活性分析

摘    要:目的:通过建立的大鼠背部囊袋金黄色葡萄球菌感染模型及无菌性炎症模型,收集炎性渗出液,并分析渗出液中各种酶的活性及其功能。方法:大鼠背部皮下注入无茵空气形成一囊袋,24 h后,对照组大鼠囊袋内注入致炎剂和生理盐水,感染组大鼠囊袋内注入致炎剂和金黄色葡萄球茵悬液,于第3,5天收集囊袋内渗出液,分析其凝血酶样活性和纤溶酶样活性。结果:与无茵性炎症比较,金葡茵感染后的渗出液中,蛋白含量明显增加,感染和非感染渗出液中分别为(48.60±7.8)rag/ml和(38.81±1.9)mg/ml;纤溶酶样活性没有明显差别;生色底物法分析感染和非感染渗出液中凝血酶样活性D405值分别为0.81±0.14和1.44±0.20。结论:研究结果表明感染性炎症比无菌性炎症创口渗出液中的蛋白含量增加,凝血酶样活性增强。

Abstract:Objectives: To experimentally establish rats models with infected or uninfected inflammation,and to evaluate the enzyme-like activity and protein contents in exudates released from wound sites. Methods: Male Wistar rats were injected with aseptic air subcutaneously to form a bursa. After 12 hours, rats of one group were injected with zymosan A and Staphlococcus aureus into the bursa(6 ?1011 cfu/L per rat) to induce inflammation. Another group was injected with zymosan A alone. Exudates in bursa of rats were collected on days 3 and 5. Protein contents and enzyme-like activity in exudates were analyzed. Results; The protein content was (48.60?7. 8) mg/ml in infected group and (38.81 ?1.9) mg/ml in uninfected group. The D405 values of thrombin-like activity in the two groups were 0.81 ?. 14 and 1.44 ?0. 20, respectively. Conclusion; The protein content in wound exudate of infected group increased more markedly than in uninfected group, Thrombin-like activity also remarkably increased in infected group as compared with the unifected group.
