吉西他滨与培美曲塞对人胰腺癌细胞BXPC-3及PANC-1生长的体外作用

目的探讨吉西他滨与培美曲塞不同用药次序联合应用对人胰腺癌细胞BXPC-3及PANC-1生长的影响及可能的细胞学机制。方法MTT法测细胞增殖;流式细胞术测细胞周期。结果吉西他滨(10-7-10mg/ml)和培美曲塞(10-7-10mg/ml)均明显抑制2株细胞生长,作用呈现浓度依赖性和时间依赖性。二者联合对2株细胞生长的影响与给药次序有关:先用培美曲塞24h后序贯用吉西他滨对2株细胞的抑制作用明显强于同时给药(P<0.05)和先用吉西他滨24h后序贯用培美曲塞(P<0.05)。吉西他滨联合培美曲塞对BXPC-3细胞周期结果显示:与对照组相比,吉西他滨与培美曲塞分别将细胞周期阻断在G1期和S期(P<0.05);同时给药G2期比例减少(P<0.05);先用吉西他滨后用培美曲塞将细胞周期阻断在G1期(P<0.05);先用培美曲塞后用吉西他滨将细胞周期阻断在S期(P<0.05)。结论吉西他滨与培美曲塞均能抑制人胰腺癌细胞BXPC-3及PANC-1生长,二者联合具协同作用,先用培美曲塞24h后用吉西他滨对2株细胞的抑制作用最强,其协同作用与他们对细胞周期的影响无关。

Effects of gemcitabine and pemetrexed on the proliferation of pancreatic cancer cell lines BXPC-3 and PANC-1 in vitro

Objective To investigate the sequence-dependent effect of combined use of gemcitabine and pemetrexed on the proliferation of human pancreatic carcinoma cell lines BXPC-3 and PANC-1 in vitro and explore the cellular mechanism.
