参芎化瘀胶囊对血管性痴呆模型大鼠海马CA_1区超微结构的影响

摘    要:目的:探讨参芎化瘀胶囊对血管性痴呆大鼠海马CA1区超微结构的影响。方法:采用反复夹闭、再通双侧颈总动脉同时ip硝普钠法制备大鼠血管性痴呆模型。SD大鼠随机分为:假手术组、痴呆模型组(模型组)、喜得镇组和参芎化瘀胶囊治疗组。以Morris水迷宫检测行为学,透射电镜观察各组大鼠海马CA1区神经元超微结构的改变。结果:①模型组与假手术组相比认知能力明显下降(P0.01),喜得镇组、参芎化瘀胶囊组与模型组相比认知能力明显改善(P0.05,P0.01)。②模型组海马CA1区神经元大量固缩,染色质凝集,异染色质边集,线粒体肿胀。喜得镇组海马CA1区神经元有少量染色质凝集,线粒体肿胀。参芎化瘀胶囊组海马CA1区多数神经元细胞核染色质分布比较均匀,胞质内线粒体及其他细胞器基本正常。结论:参芎化瘀胶囊可改善血管性痴呆模型海马CA1区损伤的超微结构,对神经元有保护作用,从而改善血管性痴呆大鼠的认知能力。

Abstract:Objective: To study the influence of Shenxiong Huayu Capsule on ultrastructures of hippocampal CA1area in rats with vascular dementia (VD). Method: The vascular dementia model of rat was established by clamping and releasing the common carotid arteries of both sides repeatly and intraperitoneal injection of sodium nitroprusside(SNP). The SD rats were evenly randomized into 4 groups: sham-operation group, VD model group, Hydergine group and Shenxiong Huayu Capsule intervention group. Cognitive ability of rats was studied with the Morris water maze. The changes in ultra-microstructure of neural cells were observed by transmission electron microscope. Result: VD model demonstrated decrease in cognitive ability (P<0.01),Shenxiong Huayu Capsule and Hydergine group alleviated cognitive ability significantly(P<0.01). In the model group, most of the neurons in CA1 region showed pyknosis, chromatin agglutination, heterochromatin margination, mitochondria swelling. In the Hydergine group, the neurons showed small amounts chromatin agglutination and mitochondria swelling. In the Shenxiong Huayu Capsule group, the distribution of chromatin was well-proportioned, and normal mitochondrion and other organelle were observed. Conclusion Shenxiong Huayu Capsule may protect neural cell through improving ultra-microstructure of hippocampal CA1area in VD Rats. Shenxiong Huayu Capsule improves cognitive ability in VD rats.
