性激素对SLE患者外周血单一核细胞凋亡的影响及其意义

摘    要:研究脱氢表雄酮(DHEA)和(或)雌二醇(E2)对系统性红斑狼疮(SLE)患者外周血单一核细胞(PBMC)凋亡的影响。应用吖啶橙(AO)染色法检测SLE患者、健康献血员PBMC与生理、病理浓度的DHEA和(或)E2培养48h的凋亡百分率。SLE患者PBMC凋亡率高于健康献血员(P<0.01)。不论生理或者病理浓度的E2对SLE患者、健康献血员PBMC凋亡无影响(P>0.05)。生理浓度的DHEA及DHEA+E2对健康献血员PBMC凋亡无影响(P>0.05),但可显著抑制SLE患者PBMC凋亡(P<0.05);病理浓度的DHEA及DHEA+E2可显著促进SLE患者、健康献血员PBMC凋亡(P<0.05)。DHEA及DHEA+E2对PBMC凋亡的影响,两者间无显著差异(P>0.05)。结果提示,DHEA对SLE患者PBMC凋亡起重要作用;DHEA浓度降低可能导致凋亡加快。

Abstract:To study effects of dehydroepiandrosterone(DHEA) and/or estradiol(E2) on apoptosis of peripheral blood mononuclear cells(PBMC) from patients with systemic lupus erythematosus(SLE), the percentage of apoptosis of PBMC from SLE patients and healthy blood donors were examined by means of AO staining 48h affter cultured with DHEA and/or E2 at physiological or pathological concentrations. The percentage of apoptosis of PBMC from SLE patients is higher than that of healthy blood donors (P< 0.01). E2, whether at physiological or at pathological concentrations,had no effects on apoptosis of PBMC from both SLE patients and healthy donors(P >0.05). Both DHEA and DHEA plus E2 at physiological concentrations, had no effects on apoptosis of PBMC from healthy donors(P >0.05),but significantly inhibited that of SLE patients(P< 0.05); while at pathological concentrations, they promoted apoptosis of PBMC from SLE patients as well as healthy blood donors(P< 0.05). There were no significant differences between the effects of DHEA and that of DHEA plus E2 (P >0.05). The results suggest that DHEA plays an important role in the apoptosis of PBMC from SLE patients; low serum levels of DHEA may cause accelerated apoptosis.
