摘    要:目的:建立HPLC法同时测定黑骨藤药材中6种咖啡酰基奎宁酸类成分的含量及液相指纹图谱,为其质量控制提供理论依据。方法:采用Agela Technologies Venusil XBP C18色谱柱(250 mm×4. 6 mm,5μm),以乙腈-0. 1%磷酸水为流动相,梯度洗脱,流速为0. 8 m L·min-1,检测波长为280 nm,柱温为25℃。结果:测定的6种成分5-O-咖啡酰基奎宁酸、3-O-咖啡酰基奎宁酸、4-O-咖啡酰基奎宁酸、3,4-O-二咖啡酰基奎宁酸、3,5-O-二咖啡酰基奎宁酸、4,5-O-二咖啡酰基奎宁酸分别在9. 84~252. 00,31. 63~506. 00,5. 76~147. 50,0. 62~15. 81,1. 45~37. 24,2. 94~75. 14 mg·L-1范围内线性关系良好。平均加样回收率分别为102. 8%,91. 3%,92. 2%,97. 4%,92. 4%,97. 4%; RSD值分别为1. 6%,2. 1%,2. 6%,2. 1%,1. 9%,1. 0%。建...

Abstract:Objective:To establish a HPLC method for the determination of six caffeoylquinic acids in Periploca forrestii and fingerprint in order to evaluate the qualitiy of Periploca forrestii.Methods:Agela Technologies Venusil XBP C18(250 mm×4.6 mm,5μm)column was adopted using the mobile phase consisted of acetonitrile-0.1%phosphoric acid in water with gradient elution at the flow rate of 0.8 m L·min^-1.The detection wavelength was set at 280 nm and the column temperature set at 25℃.Results:Six components including 5-O-caffeoylquinic acid,3-O-caffeoylquinic acid,4-O-caffeoylquinic acid,3,4-di-O-caffeoylquinic acid,3,5-di-O-caffeoylquinic acid and 4,5-di-O-caffeoylquinic acid showed good linear relationship within the ranges of 9.84~252.00,31.63~506.00,5.76~147.50,0.62~15.81,1.45~37.24 and 2.94~75.14 mg·L^-1,respectively.The average recovery of the six compounds was 102.8%,91.3%,92.2%,97.4%,92.4%,and 97.4%with the RSD value at 1.6%,2.1%,2.6%,2.1%,1.9%and 1.0%,respectively.In addition,the HPLC fingerprints of Periploca forrestii were established including 19 distinguish peaks by the developed HPLC method.And the similarity of HPLC fingerprints of 14 different batches of this plant was above 0.899.Conclusion:This method is simple,accurate and sensitive,which can be used for quality control of Periploca forrestii.
