加味佛手散抑制大鼠子宫内膜异位症血管生成及其机制

摘    要:目的探讨复方中药新药加味佛手散抑制子宫内膜异位症(EMS)血管生成作用及机制。方法采用子宫内膜自体移植法建立大鼠EMS模型,免疫组化和Western blot方法检测异位内膜组织中微血管密度(MVD)、血管内皮生长因子(VEGF)和内皮抑素(endostatin)蛋白的表达,放射免疫分析法(RIA)分别检测大鼠血清和异位内膜组织中前列腺素E2(PGE2)的含量。结果加味佛手散0.045、0.09、0.18 g.kg-1能有效抑制异位内膜组织体积的增生,加味佛手散0.09、0.18 g.kg-1能降低MVD,降低VEGF蛋白的表达,降低大鼠血清和异位内膜组织中PGE2的含量,加味佛手散0.18 g.kg-1能够升高endostatin蛋白的表达。结论加味佛手散具有抗血管生成作用,其作用机制与抑制VEGF、促进endostatin蛋白表达有关。
