摘    要:目的研究黔产粗毛淫羊藿与黔岭淫羊藿淫羊藿总黄酮(TFE)对D-gal-AlCl3联合造模大鼠学习记忆能力的影响。方法联用D-gal-AlCl3 12周造模,同时按剂量100、50 mg.(kg.d)-1,分组po给TFE。采用Morris水迷宫实验检测其空间记忆能力;应用生化技术检测脑、血AChE活性;以免疫组化技术检测脑Bcl-2/Bax水平。结果 D-gal-AlCl3造模形成了大鼠学习记忆障碍;TFE能改善D-gal-AlCl3致痴呆大鼠学习记忆能力,降低脑、血组织中AChE的活性,明显升高bcl-2并降低Bax水平。以粗毛淫羊藿改善记忆能力效果为优。结论粗毛淫羊藿对痴呆大鼠记忆能力,脑细胞凋亡具有更明显保护作用。提示淫羊藿苷(ICA)是TFE中增强记忆的主要物质基础。

Abstract:Aim To study the effects of TFE from Epimedium acuminatum and E.leptorrhizum grown in Guizhou on the learning and memory abilities of dementia model rats induced by D-gal-AlCl3 compound method.Methods The dementia model rat was induced by the D-gal-AlCl3 while 4 groups given TFE of 100,50 mg·kg-1.The Morris Water Maze was used to test their spatial learning abilities.Detect the activity of AChE in serum and brain with the biochemical method.The immunohistochemical technique was used to determine the level of Bcl-2/Bax.Results The learning and memory abilities deficit rat model was induced by the D-gal-AlCl3.The TFE improved the learning and memory abilities of the dementia rats,decreased the activity of AChE in brain and blood tissue,obviously increased the level of Bcl-2,and lowered the level of Bax.The TFE from Epimedium acuminatum was better in improving the memory abilities.Conclusions The Epimedium acuminatum is superior in enhancing the memory of dementia rats and reducing brain cell apoptosis.The results imply that ICA in TFE is the main substance for memory improvment.
