摘    要:目的:研究小鼠口腔黏膜白念珠菌感染后局部组织Toll样受体(TLR)2、4mRNA表达的变化规律,探讨其在口腔黏膜抗白念珠菌感染早期免疫中的作用。方法:采用局部接种的方法建立小鼠口腔黏膜白念珠菌感染模型。在不同时间取口腔组织.用半定量逆转录(RT)-PCR检测口腔组织TLR2、TLR4 mRNA表达水平。结果:接种前口腔组织内有微量的TLR2、TLR4 mRNA表达.接种后6h组织中TLR2、TLR4 mRNA表达开始增加,12~24h表达水平达峰值。TLR4 mRNA表达于24~48h开始下降.72h趋于接种前水平,而TLR2 mRNA表达则持续增加至72h。结论:白念珠菌感染可迅速上调局部组织TLR2、TLR4的基因表达,TLR2、TLR4在口腔黏膜抗白念珠菌感染早期免疫中可能起重要作用。

Abstract:Objective: To investigate the mRNA expression of Toll-like receptor-2 and - 4 in the oral tissues of oropharyngeal candidiasis model mice and their potential role in earlier period of immune response. Methods: A mouse model of oropharyngeal candidiasis was established by the oral inoculation of Candida albicans. Mice were sacrificed at the indicated time points and the oropharyngeal tissues were excised. Simultaneously, the expression of TLR2 and TLR4 mRNA was detected by RT-PCR. Results: The low value expression of TLR2 and TLR4 mRNA could be detected in oropharyngeal tissue before inoculation, but they were initially up-regulated 6 hours and peaked 12-24 hours after the inoculation. Then the TLR4 mRNA level was gradually reduced till to 48 hours, while TLR2 mRNA level maintained high values up to 72 hours. Conclusions: These data suggest that oropharyngeal infection of Candida albicans could result in up-regulation of TLR2 and TLR4 mRNA expressions in local tissue, which might play the important roles in the earlier period of immune reaction.
