川芎嗪对大鼠脊髓损伤后行为学改变的影响

摘    要:目的:探讨川芎嗪对大鼠脊髓损伤后行为学改变的影响。方法70只健康成年 SD 大鼠随机分为对照组( n =10)、损伤组( n =30)和药物治疗组( n =30),其中损伤组和药物治疗组接受不同处理措施后,分别于术后1、3、14 d 采用 HE 染色和电镜观察不同组大鼠脊髓形态学变化,同时采用 Jacobs 分级标准和 BBB 评分方法评价不同组大鼠后肢运动功能。结果HE 染色结果显示应用川芎嗪治疗后正常神经元数目相比较于损伤组多。电镜下可见川芎嗪治疗组神经细胞核固缩程度和染色质浓染程度较损伤组减轻,胞浆空泡化也较损伤组减轻。药物治疗14 d 组截瘫率显著低于损伤,差异有统计学意义( P <0.05)。药物治疗14 d 组 BBB 评分高于相应损伤组,差异有统计学意义( P <0.05)。结论川芎嗪治疗可改善大鼠脊髓损伤后行为学变化,对脊髓损伤有一定治疗作用。

Effects of ligustrazine on ethological changes in rats with acute spinal cord injury
Abstract:Objective To observe the effects of ligustrazine on ethological changes in rats with acute spinal cord injury.Methods Seventy healthy adult SD rats were randomly divided into control group ( n =10), spinal cord injury group (SCI group, n =30) and drug treatment group ( n =30).The rats in SCI group and drug treatment group were divided into 1-day, 3-day and 14-day subgroups after treated with different measures.The morphological changes in spinal cord of rats on1d,3d,14d after operation were detected by HE staining and electron microscopy,respectively, at the same time, the motor function in hindlimb of rats were evaluated by Jacobs classification criterion and BBB scoring .Results HE staining results showed that the number of normal motor neuron in drug treatment group was more than that in SCI group.The electron microscopy results showed that the degree of pycnosis of nerve cells,staining degree of chromatin and the vacuolization of hyalomitome in drug treatment group were obviously relieved,as compared with those in SCI group.Moreover ,the paraplegia rate on 14d after treatment in drug treatment group was 40%,which was significantly lower than that (70%) in SCI group, besides the BBB scores in drug treatment group on 14d after treatment were significantly higher than those in SCI group (10.6 ±2.7 vs 7.9 ±2.0, P <0.05).Conclusion The ligustrazine can improve ethological changes of rats after acute spinal cord injury and has certain therapeutic effects on spinal cord injury in rats.
