流式细胞术检测GDNF在胶质瘤组织中的表达

摘    要:目的:探讨胶质细胞源性神经营养因子(glial cell-derived neurotrophic factor,GDNF)表达与人脑胶质细胞瘤恶性程度的关系。方法:应用流式细胞术的方法检测GDNF在人脑胶质瘤组织中的表达水平,进行免疫荧光显微镜观察和单参数分析。结果:GDNF在人脑胶质瘤和正常脑组织中均有表达,其中阳性表达率在胶质瘤组织中分别为41.17%、41.67%、68.75%和100.00%,正常脑组织为45.00%,计算所得FI值分别为0.83-1.07、0.82-1.10、0.96-1.78和1.47-2.16,正常脑组织为0.58-0.89,显示细胞荧光强度逐渐增加。对所得数据行方差分析,各样本采用方差齐性检验,显示各样本均数差异有统计学意义,P=0.0678。样本均数间采用LSD-t检验进行两两比较显示GDNF在胶质瘤中的的表达水平显著高于正常脑组织,且随着胶质瘤恶性程度的增加表达水平也增加(P〈0.01)。结论:GDNF可能作为一种重要的因素参与胶质瘤的发生、发展及分化,其可以作为胶质瘤的检测指标,提示胶质瘤的病理分级。为进一步研究GDNF的作用及作用机制,以及探讨人脑胶质瘤的发生、发展提供了新的有意义的理论依据。

Abstract:OBJECTIVE:To study the relationship between the expression of GDNF and malignancy of human brain gliomas.METHODS:By using flow cytometry,the expressing level of GDNF in human brain gliomas was detected.The data obtained under a fluorescence microscope were processed by the one-parameter analysis.RESULTS:The level in gliomas specimens was much higher than that in NBT.The different expressions were detected in different-malignancy gliomas too:the higher malignancy,the higher level.The differentiations of positive rates(41.17%,41.67%,68.75%,100.00% and 45.00%)and FI data(0.83-1.07,0.82-1.10,0.96-1.78,1.47-2.16 and 0.58-0.89)were evident.For all gliomas and NBT the homogeneity test of variance and ANOVA was calculated,P=0.067 8.And the variance was considered significant.The pairwise comparison was analyzed by LSD-t test between the sample means(P<0.01).CONCLUSIONS:GDNF may participate in the occurrence,development and differentiation of glioma as an important factor.The results provide a reliable standard to distinguish the malignancy of gliomas or pathology classification.The results also provide a new significant theory evidence for studying the action and action mechanism,exploring the origination and development of human brain gliomas.
