微柱离心-HPLC法测定卡培他滨脂质体包封率

摘    要:采用薄膜分散法制备卡培他滨脂质体。采用葡聚糖凝胶Sephadex G-50微柱离心法分离卡培他滨脂质体和游离药物,以HPLC法测定卡培他滨浓度并计算包封率。使用C18色谱柱,以甲醇-水(55∶45)为流动相,检测波长240 nm。结果 3批脂质体的包封率分别为46.2%、45.2%和44.8%。

Abstract:The capecitabine liposomes were prepared by film dispersion method.The Sephadex G-50 gel microcolumn centrifugation method was employed to separate the free drug from the capecitabine liposomes.The capecitabine concentration was determined by HPLC to calculate the entrapment efficiency.A C18 column was used with the mobile phase of methanol-water(55∶45) at the detection wavelength of 240 nm.The results showed that the average entrapment efficiencies of three batches of capecitabine liposomes were 46.2%,45.2% and 44.8%,respectively.
