摘    要:考察了植物内生细菌欧文氏菌(Erwinia taxi)生产紫杉醇的摇瓶发酵和30 L罐发酵工艺条件。结果显示,培养基中添加不同的前体对紫杉醇产量影响显著。较高浓度的乙酸钠(>0.01 mmol/L)和较低浓度的苯丙氨酸(<0.05 mmol/L)有利于紫杉醇积累。在此基础上进行了30 L罐发酵试验,得优化培养基配方(g/L):大豆蛋白胨20,工业酵母抽提物5,氯化钠0.5,氯化钾0.1,葡萄糖4,氯化镁1,乙酸钠0.1,苯丙氨酸5×10-4;pH 6.5。发酵培养条件:接种量5%,发酵温度22℃,通气速率0.17 vvm,转速50 r/min,发酵时间60 h。在此条件下,30 L发酵罐中紫杉醇的产量为102 g/L。

Optimization of Fermentation Process of Taxol from Erwinia taxi

Abstract:A shake-flask fermentation condition of taxol from an endophytic strain of Erwinia taxi was investigated,and fermentation tests in 30 L fermentor were conducted.The results showed that addition of different precursors had significant influences on the production of taxol.Higher concentration of sodium acetate(>0.01 mmol/L) and lower concentration of phenylalanine(<0.05 mmol/L) were favourable to the taxol accumulation.The suitable contents of fermentation medium(g/L) were as follows: soya peptone 20,yeast extract 5,NaCl 0.5,KCl 0.1,glucose 4,MgCl2 1,NaOAc 0.1,phenylalanine 5×10-4 and pH 6.5 in the case of 30 L fermentor scale up.The optimized fermentation process were inoculation quantity of 5%,22 ℃,aeration rate of 0.17 vvm,stirring velocity of 50 r/min and fermentation period of 60 h.Under above conditions,the potency of taxol was tested as 102 μg/L.
