| 摘 要： | 目的 构建共刺激阻断剂CTLA4Ig基因重组的腺病毒载体。方法 将CTLA4Ig cDNA插入腺病毒表达质粒；将重组质粒与病毒基因组质粒共转染293细胞，产生重组腺病毒；采用dot-ELISA和PCR法筛选并获得CTLA4Ig阳性表达的重组腺病毒。结果 同源重组后，以dot-ELISA法筛选到2个293细胞的CTLA4Ig蛋白阳性表达空斑，PCR证实为含CTLA4Ig基因和腺病毒基因的重组体AdvCTLA4Ig。结论 本研究构建了CTLA4Ig基因重组的复制缺陷型腺病毒AdvCTLA4Ig，为进行器官移植基因治疗研究提供了工具。 |

| Abstract: | Objective To construct the CTLA4Ig gene recombined adenovidus vector.Methods The cDNA encoding CTLA4Ig was inserted into adenoviurs expression vector pAdHCMV to produce a recominant vector pAdMCMVCTLA4Ig.Subsequently, the recominant vector was cotranfected into 293 cells together with plasmid pJM17 to produce the recombinant CTLA4Ig adenovirus.Results The recombinant adenovirus AdvCTLA4Ig was generated by homologous recombinantion and was identified by dot ELISA and PCR respectively.Conclusion The constructed recombinant AdvCTLA4Ig adenovirus vector provides a potent tool for gene therapy in the induction of immune tolerance in organ transplantation. |
