变形链球菌葡糖基转移酶基因在戈登链球菌中的表达

摘    要:目的:通过探讨变形链球菌葡糖基转移酶基因在戈登链球菌中的表达,了解葡糖基转移酶对戈登链球菌生物学特性的影响。方法:携带的大肠杆菌链球菌穿梭质粒pZB1转化戈登链球菌,利用蛋白质免疫印迹技术确认葡糖基转移酶基因在转化株中表达,通过检测转化株菌落形态以及蔗糖依赖性粘附了解葡糖基转移酶对戈登链球菌生化特性的影响。结果:戈登链球菌的菌落形态由转化前的光滑型变为转化后的半粗糙型,转化菌株的蔗糖依赖性粘附较转化前明显降低。结论:变形链球菌的葡糖基转移酶在戈登链球菌中的表达增加了细菌之间的聚集,葡糖基转移酶与戈登链球菌的粘附因子相互作用,降低了细菌的蔗糖依赖性粘附。

Abstract:Objective: To investigate the influences of Streptococcus mutans glucosyltransferase-B gtfB on the biochemical characteristics of streptococcus gordonii. Methods: E.coli-streptococcus shuttle plasmid pZB1 containing streptococcus mutans gtfB was transformed into streptococcus gordonii. Western blotting was used to determine whether gtfB was expressed in the transformant. The colony morphologies and in vitro sucrose-dependent adherence were detected to investigate the biochemical characters of streptococcus gordonii before and after being transformed.Results: The colony morphologies of streptococcus gordonii became smaller than the wild type. Western blotting results showed that one protein of the mutant reacting with the antibody against streptococcus mutans glucosyltransferase-B glucan-binding fragment had the same molecular weight as that of streptococcus mutans MT8148 cell-associated glucosyltransferase. In vitro sucrose-dependent adherence of streptococcus gordonii decreased significantly after being transformed. Conclusion: The expression of streptococcus mutans gtfB in streptococcus gordonii increased coaggregation of the cells.
