神经生长因子对腺样囊性癌神经侵袭作用的实验研究

摘    要:目的:探讨神经生长因子(nervegrowthfactor,NGF)对腺样囊性癌(adenoidcysticcarcinoma,ACC)神经侵袭的作用。方法:免疫组化方法检测8只神经侵袭裸鼠模型体内ACC癌组织中NGF的表达,比较邻近神经的ACC癌组织与远离神经的癌组织中NGF阳性表达细胞的密度。另10只神经侵袭模型的裸鼠随机分为实验组和对照组,实验组裸鼠腹腔内注射抗NGF抗体40μg/次,隔天一次,持续14d;对照组注射等量磷酸盐缓冲液。统计学t检验比较两组神经侵袭长度的差异。结果:ACC细胞中表达NGF,且邻近神经的ACC中NGF阳性表达细胞的密度显著高于远离神经的ACC。体内注射NGF抗体的实验组动物神经侵袭的长度为(0.156±0.025)mm,显著短于对照组的(0.378±0.050)mm,P<0.01。结论:ACC中NGF的表达增强与神经侵袭相关,NGF能促进ACC的神经侵袭。

Abstract:Objective: To investigate the role of nerve growth factor (NGF) in neural invasion of adenoid cystic carcinoma(ACC). Methods: Expression of NGF was detected in ACC cells by immunohistochemistry in 8 nude mice models of neural invasion, and density of NGF positive cells in ACC adjacent to nerve and that of ACC far from nerve was compared. Other 10 nude mice models of neural invasion were randomly divided into experimental and control groups. Intra peritoneal injection of 40 μg anti-NGF antibody was performed at two-day intervals for 14 days in experiment mice, and phosphate buffer solution was applied in control mice. Neural invasion length was measured 14 days after injection and difference between experimental and control group was analyzed by T-test. Results: NGF was expressed in ACC cells. Density of NGF positive cells in ACC adjacent to nerve was significantly stronger than that of ACC cells far from nerve in animal model. The length of neural invasion in experimental group(0.156±0.025) mm] was markedly shorter than that in control group(0.378±0.050) mm], P<0.01. Conclusion: The density of NGF expression in ACC was correlated with the degree of neural invasion. As neutralizing NGF, anti-NGF antibody can suppress neural invasion of ACC, NGF can promote neural invasion of ACC.
